Improving treatment outcomes
Insulin can increase the cytotoxic effect of methotrexate up to 10,000-fold in MCF-7 human breast cancer cells in vitro.1Alabaster O, Vonderhaar BK, Shafie SM. Metabolic modification by insulin enhances methotrexate cytotoxicity in MCF-7 human breast cancer cells. European Journal of Cancer and Clinical Oncology. 1981 Nov;17(11):1223-8.
Insulin-pretreated colon cancer cells are significantly more susceptible to commonly used chemotherapeutics.2Agrawal S, Woźniak M et al. Insulin enhancement of the antitumor activity of chemotherapeutic agents in colorectal cancer is linked with downregulating PIK3CA and GRB2. Scientific Reports. 2019 Nov 12;9(1):16647.
“In 14 cell lines and 2 animal studies, chemotherapy response was lower in a hyperglycemic (greater than 15 mmol/L) compared to a euglycemic environment (5 mmol/L). The response was similar in 4 cell lines. In the remaining 5 cell lines, the hyperglycemic environment potentiated chemotherapy efficacy.”3Gerards MC, van der Velden DL et al. Impact of hyperglycemia on the efficacy of chemotherapy—a systematic review of preclinical studies. Critical Reviews in Oncology and Hematology. 2017 May;113:235-241.
